Title |
Microglia_specificpathogenfree_4d_after_LCMV_ic_01 |
Sample type |
RNA |
Source name |
Microglia, LCMV i.c., SPF
Organism |
Mus musculus |
Characteristics |
treatment: LCMV condition: specifc pathogen free (SPF) cell type: microglia
Treatment protocol |
Lymphocytic Choriomeningitis Virus (LCMV) strain WE was propagated and titrated as plaque forming units (PFU) on L929 cells as described before 12. PFU were multiplied by the factor 10 to be converted into infectious units (IU). Mice were infected by intracerebral inoculation of 103 IU into the right hemisphere
Growth protocol |
Mice were housed under SPF or germ-free condiotions. Animals were 6-10 weeks old.
Extracted molecule |
total RNA |
Extraction protocol |
Adult microglia were harvested using density gradient separation. CD11b+CD45lo microglial cell populations were FACS-sorted and collected directly in cell lysis buffer and subsequently RNA was isolated with the Arcturus Pico Pure RNA Isolation Kit (Life Technologies) according to the manufacturer's protocol.
Label |
Biotin
Label protocol |
Total RNA was amplified using the Ovation PicoSL WTA System V2 and further processed using the Encore Biotin Module according to the instructions supplied by the manufacturer (NuGen).
Hybridization protocol |
Biotinylated cRNA was hybridized on GeneChip® Mouse Gene 2.1 ST Arrays , that were stained, washed and scanned following standard procedures.

Description |
Microglia from specifc pathogen free (SPF) mice treated with LCMV i.c.
Data processing |
Background correction and normalization was performed using the RMA-Sketch implementation in Affymetrix Expression Console
