in vitro Th17 cells GFP sorted
Organism |
Mus musculus |
Characteristics |
cell type: in vitro differentiated Th17 genotype: OT-II IL17GFP knock-in
Treatment protocol |
None
Growth protocol |
cells derived from OT-II IL17GFP knock-in were differentiated into Th17 in vitro by culturing splenocytes with IL-1b (10 ng/mL), IL-6 (10 ng/mL), IL-23 (10 ng/mL), human TGF-b (2.5 ng/mL) for 4 days and OVA 323-339 peptide or by in vivo exposure to Freund’s Adjuvant and OVA 323-339 peptide for 7 days
Extracted molecule |
total RNA |
Extraction protocol |
RNA was extracted with RNease Plus Mini Kit (Qiagen) and samples were processed and analyzed at the University of Michigan microarray facility with Affimetrix mouse Gene 1.0 ST array.
Label |
biotin
Label protocol |
cDNA were prepared according to the NuGen WT-Pico V2 kit protocol from 50 ng total RNA (Ovation PicoSL WTA System V2 P/N 3312). Biotinylated single-stranded cDNA were prepared from 3ug of cDNA (Encore Biotin Module P/N 4200-12, 4200-60, 4200-A01)

Hybridization protocol |
Following fragmentation, 3.7 ug of cDNA were hybridized for 20 hr at 48C on House Gene ST 2.1 strip arrays using the Affymetrix Gene Atlas system (software version 1.0.4.267).
Scan protocol |
Arrays were scanned using the Affymetrix Gene Atlas system.
Description |
Gene expression data from in vitro Th17 cells
Data processing |
Data was analyzed using the oligo Bioconductor package implemented in the R-statistical environment. RMA was used to fit log2 transformed expression values based on a larger set of samples, which have not been deposited.
