Extraction protocol |
Total RNA (including microRNAs) was extracted from frozen tissues using the TRIZOL reagent (Invitrogen) following the manufacturer’s recommended protocol. Concentration and purity of the total RNAs were assessed by spectrophotometer (Nanodrop 1000, Nanodrop Technologies, Wilmington, DE, USA) and RNA integrity was verified using an Agilent 2100 bioanalyzer (Agilent Technologies).

Cy3-labeled RNA was generated using the Agilent’s miRNA Complete Labeling and Hyb Kit with a sample input of 100 ng of total RNA.

Hybridization protocol |
Hybridization of labeled miRNA to 8x15K Rat miRNA microarray slides (based on miRBase release 16.0) was for 20 hours at 55ºC on a rotator at 20 rpm.

Scan protocol |
The slides were then washed and scanned with an Agilent microarray scanner.
