Title cardiomyocytes 5 day after proton radiation exposure rep2
Sample type RNA

Source name left ventricular murine cardiomyocytes to 90 cGy, 1 GeV proton (1H) 5 day after exposure
Organism Mus musculus
Characteristics tissue: left ventricular murine cardiomyocytes
strain: C57Bl/6NT
Sex: male
age: adult (8-9 months old)
Treatment protocol Irradiation and Dosimetry – Whole body irradiation of adult (8-9 months old) male C57Bl/6NT (Taconic) mice was performed at the NASA Space Radiation Laboratory (NSRL) at Brookhaven National Laboratory (Upton, NY). Using dose-rates between 0.05 Gy/min and 0.17 Gy/min, iron and proton, respectively; 0.15 Gy was delivered using 1 GeV/nucleon iron ions (LET ~151 keV per µm) or 0.9 Gy at 1 GeV energy protons (LET ~0.22 keV per µm).
Extracted molecule total RNA
Extraction protocol Cardiac Cell Isolation – Primary left ventricular cardiac cells from 8-9 months old C57Bl/6NT mice were isolated using standard preparation protocol that includes cannulation of the aorta and collagenase digestion followed by a Ca2+ gradient selection (22, 25, 26, 33). This procedure yielded >90% of Ca2+ tolerant cardiomyocytes (CMs). The resulting cell suspension could contain a small population of inflammatory and/or endothelial cells. Following isolation of CMs, RNA was extracted via TRIzol (Life Technologies, Grand Island, NY) according to the manufactures instructions. RNA quality and quantity was assessed using the 2100 Bioanalyzer (Agilent Tech., Santa Clara, CA).
Label biotin
Label protocol Biotinylated cRNA were prepared according to the standard Affymetrix protocol.

Hybridization protocol Follow manufacturor Affymetrix standard procedure
Scan protocol Scanned with Affymetrix GeneChip imager
Data processing MAS5.0 signal intensities analyzed using Dchip in Webarray
