Growth protocol |
Animals were kept in a SPF barrier under standardisized conditions with respect to temperature and humidity, and were housed on a 12h light/12h dark cycle in groups of 4-5 with free access to food and water. Animal housing, care, and applications of experimental procedures complied with the Guide for the Care and Use of Laboratory Animals of the Government of Berlin, Germany and are in accordance to the recommendations of the Society for Laboratory Animal Science (GV-SOLAS) and the Federation of European Laboratory Animal Science Associations (FELASA).

ErbB4 knockout mice were treated 3 times a week with 15mg/kg Doxorubicin.

Hybridization protocol |
Targets were vacuum-centrifuged and resuspended in 50 μl DIG hybridisation buffer (Roche). Denatured targets (98°C, 2 min) were hybridised in a humidified chamber at 42°C overnight. The following day arrays were washed (2xSSC; 0,03% SDS for 10min, 2xSSC for 5min, 1xSSC for 5min, 0,2 x SSC for 2min) and dried in a centrifuge.

The gene expression of the whole heart of ErbB4 knockout mice treated with doxorubicin was compared with wildtype mice.

Data processing |
Local background was subtracted and LOWESS normalisation was performed using Feature Extraction Software (version A 6.1.1).
