Treatment protocol Cells were infected with retoviral vectors (empty or miR-34a) and selected for 2 weeks in 1 ug/mL puromycin.
Growth protocol HCT116 cells were grown in McCoy's 5A media plus 10% fetal bovine serum plus antibiotics.
Extracted molecule total RNA
Extraction protocol Total RNA was extracted using the Qiagen RNeasy kit according to the manufacturer's instructions.
Label biotin
Label protocol Biotinylated cRNA were prepared according to the standard Affymetrix protocol from 5 ug total RNA (Expression Analysis Technical Manual, 2001, Affymetrix).
 
Hybridization protocol Following fragmentation, 10 ug of cRNA were hybridized for 16 hr at 45C on Affymetrix U133 Plus 2.0 arrays. GeneChips were washed and stained using the Affymetrix Fluidics Station 400.
Scan protocol GeneChips were scanned using a Hewlett-Packard GeneArray Scanner G2500.

Chang TC, Wentzel EA, Kent OA, Ramachandran K, Mullendore M, Lee KH, Feldmann G, Yamakuchi M, Ferlito M, Lowenstein CJ, Arking DE, Beer MA, Maitra A, and Mendell JT. (2007). Transactivation of miR-34a by p53 broadly influences gene expression and promotes apoptosis. Mol Cell, 26: 745-752.
Data processing The data were analyzed with Microarray Suite version 5.0 (MAS 5.0) using Affymetrix default analysis settings and global scaling as normalization method. The trimmed mean target intensity of each array was arbitrarily set to 150.
