Cells were pelleted at 350g and resuspended in 350ul of RLT buffer (Qiagen) and frozen at -80°C.

Growth protocol |
For colony-forming assays MethoCult M3236 (Stem Cell Technologies) was used and supplemented with 5U/mL rhEPO, 50ng/mL mSCF, 100nM dexamethasone and 1% antibiotics. Cells were resuspended in FCS and added to MethoCult mixture. Colonies were scored at day 8.
