Growth protocol mES cells were routinely generated by re-plating 3-4 days onto fresh MEF-seed dishes in mESC growth medium that consisted of DMEM with high glucose (Hyclone) supplemented with 20% Knockout serum replacement and 1000 U/ml LIF.

Extraction protocol Total RNA was isolated using TRIzol® RNA Isolation Reagents (Life technologies, Carlsbad, CA)
The isolated total RNA was processed for preparing mRNA sequencing library using TruSeq stranded mRNA sample preparation kit (illumina, San Diego, CA) according to manufacturer’s instruction.
