Sample GSM1976925 Query DataSets for GSM1976925
Status Public on Dec 19, 2016
Title WT_heart_isoproterenol
Sample type SRA

Source name heart ventricles_WT_isoproterenol
Organism Mus musculus
Characteristics tissue: heart ventricles
developmental stage: 8-12 weeks old mice
background strain: C57Bl6/J
genotype: WT
treatment: isoproterenol
Growth protocol WT and P2Y6 KO mice aged between 8 and 12 weeks were intraperitoneally injected with 50 mg/kg/day isoproterenol (Sigma Aldrich) or saline solution, daily during 7 days.
Extracted molecule total RNA
Extraction protocol Mice were sacrificed and hearts were harvested and weighed. Ventricles were then stored in TrizolĀ® reagent (Life Technologies) for RNA extraction
Indexed cDNA libraries were obtained using the TruSeq Stranded mRNA sample preparation kit (Illumina) following manufacturer recommendation.

Library strategy RNA-Seq
Library source transcriptomic
Library selection cDNA
Instrument model Illumina HiSeq 1500

Data processing Approximately 25 million paired-end reads (2x100) /sample were collected on a Hiseq1500 sequencing system (Illumina)
Paired-end reads (cut at 1-85 bases) were mapped against the mouse reference genome (GRCm38.p4/mm10) using STAR2.5 software to generate read alignments for each sample. Annotations Mus_musculus.GRCm38.82.gtf were obtained from ftp.Ensembl.org.
After transcripts assembling, gene level counts were obtained using HTSeq, followed by a library size normalization
Genome_build: GRCm38.p4/mm10
Supplementary_files_format_and_content: CSV file include CPM (count per million) values for each Sample.
