Extraction protocol |
Samples were taken from epididymal fat and frozen on dry ice. Frozen tissues were individually pulverized. Total RNA was extracted from the resultant frozen tissue powder using TRI-reagent (Sigma) according to the manufacturer's recommendations. Extracted RNA was additionally purified on RNeasy mini columns (Qiagen). RNA quantity and quality was verified on Bioanalyser 2100 (Agilent). For each strain, RNA was pooled so that the final amount was 3 micrograms total RNA, with each mouse represented in equal proportions. The number of mice pooled per strain is noted under the Sample Description section.
