Total RNA was extracted from 1.5 - 2.0 mL serum using Plasma/Serum Circulating and Exosomal RNA Purification Mini Kit (Cat # 51000) according to manufacturer’s instructions (Norgen Biotek, Canada).

In labeling reaction, 400 - 600 ng of total RNAs were used for reverse transcription using GoScript™ Reverse Transcription system (Promega, USA) with random primers and Cyanine-5-dUTP (Cat # ENZ-42502, Enzo Life Sciences, USA). Labeled RNAs were purified using Micro Bio-Spin 30 column (Bio-Rad, USA).

Hybridization protocol |
The labeled RNAs were hybridized to the array with a 1×hybridization solution (5× Denhart’s solution, 0.5% SDS, 5× SSC) in a Hybridization Chamber (Corning Inc, Corning, NY, USA) at 45°C for 12 to 18 hours.

Scanned in a LuxScanTM -10K Scanner (capitalBio Inc.,Beijing, China).

Data processing |
Firstly, we gridded the microarray image using GenPix Pro 6.0 program, and subtracted background and normalized the data (quantile normalization) using GPR analysis software(edited in-house). Then we computed the average intensity of the duplicated probes and transformed them into log2 value.
