Growth protocol Yeast cells were grown in YPD medium supplemented with/without 4mM H2O2
Extracted molecule total RNA
Extraction protocol Total RNA was extracted with RNeasy® Mini Kit and RNase-Free DNase Set following manufacture's instructions. RNA quality and integrity was verified by gel electrophoresis, as well as by measuring 260/230 ratios with a NanoDrop 1000 spectrophotometer (Thermo Scientific, USA).
Label Cy3
Label protocol Cyanine-3 (Cy3) labeled cRNA was prepared from 0.5 ug RNA using Low Input Quick Amp Labeling Kit, One-Color (Agilent) according to the manufacturer's instructions
 
Hybridization protocol 1.5 ug of Cy3-labelled cRNA (specific activity >10.0 pmol Cy3/ug cRNA) was applied for the hybridization process. To ensure the accuracy of the collected data, Agilent G2545A Hybridization Calibration Procedure (p/n G2545-90002, version A1 or higher) was performed.
Scan protocol Slides were scanned immediately after washing on SureScan microarray scanner using one color scan setting for 8x15k array slides
Description Gene expression when OD600 of cell culture reached 1.0
Data processing Agilent Genespring GX software was used for raw data analysis and p-values were calculated with unpaired Student t-test
Normalized signal intensity, using the 75th percentile signal value for inter-array comparisons. Compared to the median or 50th percentile signalsignal value, the 75th percentile of all of non-control probes on the microarray is a more robust and representative value of the overall microarray signal.
