Title |
R+SAHA_3 |
Sample type |
RNA |
Source name |
R+SAHA_3
Organism |
Manihot esculenta |
Characteristics |
authority: Manihot esculenta Crantz cultivar: TMS6044 tissue: Root salt condition: unstress condition saha condition: with SAHA
Treatment protocol |
The plantlets showing root elongation were treated with 100 µM SAHA for 24 h, followed by treatment with 200 mM NaCl for 2 and 24 h.
Growth protocol |
1-week old stem cut cassava TMS6044 were grown on liquid MS medium (no agar).
Extracted molecule |
total RNA |
Extraction protocol |
Total RNA was extracted using Plant RNA Isolation Reagent (Invitrogen) following modified spin-column RNA precipitation using RNeasy Plant Mini Kit (Qiagen).
Label |
Cy3
Label protocol |
100 ng of total RNA were used to prepare for Cy3-labeled cRNA, using Low Input Quick Amp Labeling Kit (Agilent).
Hybridization protocol |
600 ng of cRNAs from total RNA samples were hybridized at 65 degree for 17 hours, using Gene Expression Hybridization kit. After hybridization, the microarray was washed with Gene Expression Wash Buffer 1 (Agilent) and Gene Expression Wash Buffer (Agilent), and then dried immediately by brief centrifugation.
Scan protocol |
The microarray was scanned by Agilent DNA Microarray Scanner G2539A ver. C and raw data was extracted using Feature Extraction program ver 9.1.
Data processing |
RMA normalization was performed for signals of microarray probes using limma package on R program.
