Extraction protocol |
Spleen were removed, stained TH1 cells or TFH cells were sorted using flow cytometer, and RNA was harvested using Trizol reagent. RNA sequencing libraries were prepared using a kit from Illumina (TruSeq sample prep kit v2; for samples 1-7) or New England Biolabs (NEBNext Ultra RNA Library Prep kit; the rest of samples) RNA sequencing was done with an Illumina HiSeq1500 on a 50-base single-end mode.
