Treatment protocol The cell lines Bel-7402 was seeded in 6-well plate at a concentration of 2×10^5 per well and incubated overnight. Then the cells was treated with various concentration (0,40 and 80μM) of baicalein dissolved in DMSO (final DMSO concentration in growth media, 0.1%) for 24h.
Growth protocol Human hepatocellular carcinoma cell lines Bel-7402 was cultured in Dulbecco’s modified Eagle’s medium (Hyclone, USA) supplemented with 10% fetal bovine serum (Gibco, USA), 1% penicillin-streptomycin (100 U/ml penicillin and 100 μg/ml streptomycin; Hyclone, USA) at 37°C in a humidified atmosphere with 5% CO2.
Extracted molecule total RNA
Extraction protocol After a 24h incubation, the cell were harvested and the total RNA including miRNAs was extracted using TRIzol reagent (Invitrogen, USA) according to the manufacturers’ instructions.
Label biotin
Label protocol Per RNA sample, 1 μg of total RNA was labeled with biotin using FlashTag® Biotin HSR RNA Labeling Kit (Genisphere, USA) according to the manufacturer's instructions.
 
Hybridization protocol Labeled miRNA was hybridized to the array for 16 hours at 48 “C and 60 rpm and were then washed and stained using the GeneChip ® Hybridization Wash and Stain Kit in the 450 Fluidics Station according to the manufacturer's instructions.
Scan protocol GeneChips were scanned using the Hewlett-Packard GeneArray Scanner G3000 7G.
Data processing The raw data (CEL files) were extracted from the scanned images using the Affymetrix GeneChip Command Console. Expression data were generated by Affymetrix Expression Console software and normalized by the MAS5 method
