Extraction protocol PBMCs were purified from 60 ml blood by Ficoll density gradient centrifugation (U16 NovaMed). The CD14 subset was isolated using EasySep™ Human Monocyte Enrichment Kit (#19059 Stemcell). Recovery was 65-80% with a purity > 95% for all cell-subsets. From each sample both RNA and DNA was extracted using AllPrep kit (#80204 QIAGENE) according to manufacturer's protocol. DNA concentration was measured by picogreen and stored at 4°C until use. RNA quantity and quality was measured by NanoDrop and Agilent BioAnalyzer respectively and stored at -80°C until use.
