Treatment protocol Cells were seeded 37.500 cells/cm2, cultured for 24 hours and transfected with siRNA (scrambled or targeting Notch1). 48 hours after transfection, cells were stimulated with recombinant human interleukin-1b (5ng/ml, control cells were given fresh medium) for 4 hours before harvesting in Qiagen RLT lysis buffer.
Growth protocol Pooled HUVEC from 10 donors were cultured in MCDB 131 with 7.5% FBS, 10 ng/ml EGF, 1 ng/ml bFGF, 1μg/ml hydrocortisone, 50 μg/ml gentamicin, 250 ng/ml fungizone and 1% L-glutamine. Cells were maintained at 37°C in human 95% air/ 5% CO2 atmosphere.
Extracted molecule total RNA
Extraction protocol RNA was extracted using the Qiagen RNEasy Mini kit, according to manufacturers instructions.
