Treatment protocol |
Mice received either (a) a standard rodent chow diet (A03-10 UAR, control) (CHOW), (b) a standard rodent chow diet and metformin (0.5 g/l in drinking water, ca 50–80 mpk/d) (MET), (c) a standard rodent chow diet containing 0.5% (wt/wt) TCA (TCA), or (d) a standard rodent chow diet containing 0.5% (wt/wt) TCA and metformin for 5 days (TCA+MET). Mice were sacrificed by cervical dislocation after 6 hours of fasting.

Growth protocol |
C57BL6/J mice were purchased from Charles River Laboratories and housed in a temperature-controlled room (22–24°C) with a relative humidity of 36%–80%, and 12-hour light/12-hour dark cycles. Mice were fed ad libitum, with free access to water.

ChIP was performed as described in Flajollet,S., Lefebvre,B., Rachez,C., and Lefebvre,P. 2006. Distinct Roles of the Steroid Receptor Coactivator 1 and of MED1 in Retinoid-induced Transcription and Cellular Differentiation. J. Biol. Chem. 281:20338-20348. Libraries were prepared using NEBNext® ChIP-Seq Library Prep Master Mix Set for Illumina (E6240 and E7335 from New England Biolabs) according to the manufacturer's instructions

Reads were mapped to the mouse genome (mm10) using Bowtie 1 Bigwig files report signal intensity at 25 bp resolution normalized to the library size. Genome_build: mm10 Supplementary_files_format_and_content: Bigwig (signal intensity at 25 bp resolution normalized to the library size).
