Treatment protocol Temperature in the experimental house was increased each day from 25 degrees celsius to approximately 35-37 degrees celsius from 7 am to 3 pm each day starting on day 21. This cyclical heat stress scheme continued through day 42, completion of the trial.
Growth protocol Birds were raised following grower standards outlined by our AACUC protocol (#(27)03-12-14R). Briefly, birds were raised on litter with ad libitum access to feed and water. First week house temperature: 35 degrees C with 5 degrees C decrease each week until 25 degrees celsius was reached. Thermoneutral/Control house maintained this temperature through completion of the trial while heat stress (experimental) house began a cyclical heat stress scheme detaield below through completion of the trial at day 42 (typical market age). Both houses received 23 hours of light and 1 hour of dark for the entirety of the study. Necropsies were performed on Day 21 (prior to treatment), Day 22 (8 hours of treatment), Day 26 (2 hours acute treatment), Day 32 (4 hours acute treatment) and Day 42 (3 weeks of cyclical heat stress). Matched controls were collected at each time point.

Extraction protocol Birds were euthanized via cervical dislocation and necropsied on each day listed above. Whole pituitary glands were collected and flash frozen with liquid nitrogen.
Libraries were constructed using the Illumina Stranded RNA-seq kit according to manufacturers instructions.
