Treatment protocol |
Cells were grown to OD600nm = 0.6 and then treated with benomyl (Sigma-Aldrich) to a final concentration of 20μg/ml (stock solution: 10 mg/ml in DMSO). The mock treatment was done with an equivalent volume of DMSO. The cells were snapped- frozen in cold ethanol (final concentration 70% at -80°C) after 2, 4, 10, 20, 40 and 80 minutes of benomyl or mock treatment.

All experiments were conducted in YPD rich media (2% glucose, 1% bactopeptone, 1% yeast extract).

Artefactual and saturated signal spots were eliminated. After image quantification, data were normalized over all features with print-tip lowess using the R/BioConductor packages “limma” and “marray” available at http://www.bioconductor.org/.
