Treatment protocol In vitro whole blood infection was performed using different strains of ZIKV. At 40 hpi, peripheral blood mononuclear cells (PBMCs) were isolated using density gradient centrifugation using SepMate and lymphocyte separation medium. Total monocytes were further isolated from PBMCs using Pan Monocyte Isolation Kit.

Extraction protocol 10,000 CD14+ sorted monocytes were lysed in RLT buffer at a concentration of 6500 cells/ul.

Scan protocol Sample Cartridges are placed in the Digital Analyzer for data collection. Color codes on the surface of the cartridge are counted and tabulated for each target molecule. The data produced by the nCounter Digital Analyzer are exported as a Reporter Code Count (RCC) file.
Data processing Data collection is performed using epifluorescence microscopy and CCD capture technology on an nCounter instrument to yield hundreds of thousands of target molecule counts. Digital images are processed within the nCounter instrument, and the Reporter Probe counts are tabulated in a comma-separated value (CSV) format or RCC file for downstream data analysis with NanoString’s nSolverTM Analysis Software.
