RNA was isolated with Trizol(r) method according to the manufacturer's protocol. The RNA was purified with the RNeasy MiniElute Cleanup Kit (Qiagen, Hilden, Germany)

50 to 100 nanograms of total RNA was used to generate double-stranded cDNA with a T7(dT)24-oligonucleotide primer performed with the One Cycle cDNA Synthesis Kit (Affymetrix, Santa Clara, CA, USA) followed by a second IVT reaction to amplify the cDNA. After purification with the Sample Cleanup Module (Affymetrix), cDNA served as a template to prepare biotinylated cRNA via IVT, using the IVT Labeling Kit (Affymetrix). The labeled cRNA transcripts were purified using the Sample Cleanup Module.
