NCI-H209 cells (1 × 107) were mixed with 1 μM siRNAs in an electroporation cuvette and electroporated using Gene Pulser II Electroporation Systems (Bio-Rad, Hercules, CA, USA) with voltage 200 V and capacitance 950 μF.

NCI-H209 cells were cultured in RPMI1640 + 10% FBS.

Extraction protocol |
Total RNA was extracted 48 h after siNC or siTTF-1 transfection, using RNeasy Mini Kit (Qiagen, Hilden, Germany). Total RNA QC was performed by Agilent 2100 Bioanalyzer.

Biotin-labeled samples were prepared using FlashTagTM Biotin HSR RNA Labeling Kit for Affymetrix GeneChip miRNA Arrays (Affymetrix, Inc., cat.# 901911) .

Hybridization protocol |
Hybridization was performed with Hybridization Control Kit (Affymetrix, Inc., cat.# 900454) and Hybridization, Wash, and Stain Kit (Affymetrix, Inc., cat.# 900720) in GeneChip Hybridization Oven 645 (Affymetrix, Inc.) for 18 hr (60 rpm) at 48 degree celsius. Washing was performed using GeneChip Fluidics Station 450 (Affymetrix, Inc.).
