Treatment protocol For Recombination Induced Tag Exchange experiment (RITE), cells were treated with 1 mM b-estradiol.
Growth protocol Schizosaccharomyces pombe cell were gwon in rich medium (1X or 4X YES). Cell cycle synchronisation were performed using cdc25-22 temperature dependent block release.
Extracted molecule genomic DNA
Extraction protocol Lysates were prepared using standard bead beating procedure, sonicated to achieve fragmements of 200-400bp andclarified by centrifugation at 17000g.
Libraries were prepared using standard Illumina pipeline. Briefly, DNA was end-repaired using NEB Quick blunting kit (E1201L). The blunt, phosphorylated ends were treated with Klenow fragment (3' to 5' exo minus) and dATP to yield a protruding 3- 'A' base for ligation of NEXTflex adapters (Bioo Scientific) which have a single 'T' base overhang at the 3’ end. After adapter ligation DNA was PCR amplified with Illumina primers for 13-15 cycles and library fragments of ~300 bp (insert plus adaptor and PCR primer sequences) were selected using Ampure XP beads. The purified DNA was captured on an Illumina flow cell for cluster generation.
