Growth protocol |
Balb/c mice were challenged with 5E5 C26GM cells sc. 9 days later, CD11b+ cells were magnetically isolated (Miltenyi) from RBC lysed splenocytes or collagenase (Sigma) treated tumors. CD11b+ cells purity was higher than 95% as per flow cytometry analysis.

Extraction protocol |
Trizol extraction of total RNA was performed according to manufacturer guidelines, with minor modifications. Extraction was followed by Qiagen RNeasy column clean-up according to manufacturer instructions.

RMA of .CEL files using Bioconductor affy library and Brainarray custom CDF Mouse4302_Mm_ENTREZG_21.0.0. In RMA, PM values have been background adjusted, normalized using quantile normalization, and expression measure calculated using median polish summarization.
