Sample GSM3031682 Query DataSets for GSM3031682
Status Public on Jan 17, 2019
Title normal control, biological rep 2 (STZ-diabetic study)
Sample type RNA

Source name Whole retina from normoglycemic animals @6 weeks
Organism Mus musculus
Characteristics background strain: C57Bl6/J
tissue: Whole retina
disease state: normoglycemic
streptozotocin: no
Treatment protocol Enucleated Eyes were snap frozen in liquid nitrogen and stored at -80°C till further preparation. Extraction of the retina was performed at 0°C and immediately suspended in Trizol.
Growth protocol Diabetes was induced by intra peritoneal injection of streptozotocin (160 mg/kg/bodyweight; Sigma) in 8-week-old outbred male C57BL6/J mice weighing 24-26 g. 7 days after injection, stable hyperglycemia was confirmed (blood glucose >15 mM). Non-diabetic animals served as controls. Body weight and blood glucose values were monitored at regular intervals. Animals were housed with a 12-/12-h light/dark cycle with ad libitum access to standard rodent chow. Termination occured at 6 weeks.
Extracted molecule total RNA
Extraction protocol Trizol extraction of total RNA was performed according to the manufacturer's instructions.
Label biotin
Label protocol Biotinylated cRNA were prepared according to the standard Affymetrix protocol.

Hybridization protocol Hybridization (16h x 45°C) was processed according to the standard Affymetrix protocol.
Scan protocol Affymetrix GeneArray Scanner3000
Description Gene expression data from normal control C57Bl6/J mice after 6 weeks
NC_6W_1172
Data processing The data were analyzed with a commercial software called JMP Genomics, version 6, from SAS. Gene expression profiling was performed using arrays of mouse Mogene-2_0-type from Affymetrix. A Custom CDF Version 19 with Entrez based gene definitions was used to annotate the arrays. The Raw fluorescence intensity values were normalized applying quantile normalization, RMA background correction and Medianpolish Probeset Summary
