Data processing |
R package "fields" was used to normalize hybridisation data. Initially, a smooth three dimensional surface was fitted to the data. For each individual spot the height of the surface at the position of that spot is then subtracted from its raw signal intensity value. The height of this surface at a specific position represents the local average signal. In order to avoid negative values the overall mean of the smooth fit is added to all signal intensities.
