Title |
WT L4 B: Lactation day 4, rep 2 |
Sample type |
RNA |
Source name |
Mammary Epithelial Cells, depleted of mammary adipocytes, at lactation day 4
Organism |
Mus musculus |
Characteristics |
strain: FVB Sex: female tissue: Mammary Epithelial Cells depleted of mammary adipocytes parity: 1st stage of mammary development: Secretory Activation
Treatment protocol |
Mammary Epithelial Cells (MEC) at pregnant day 14 and lactation day 4 were depleted of the mammary adipocytes by rapid enzymatic tissue digestion (PMID: 20739508), washed with PBS, pelleted, and snap frozen for total RNA expraction.
Growth protocol |
FVB mothers were staged for pregnancy by post coital plug and sacrificed at either day 14 of pregnancy or day 4 of lactation (day 1 is the first morning litters are born).
Extracted molecule |
total RNA |
Extraction protocol |
Trizol extraction of total RNA was performed according to the manufacturer's instructions. Total RNA was cleaned up and DNA was digested on column according to the Qiagen Rneasy mini kit.
Label |
Biotin
Label protocol |
Biotinylated sense-strand DNA targets from total RNA were generated according to the Affymetrix WT PLUS Reagent Kit for GeneChip® Whole Transcript (WT) Expression Arrays
Hybridization protocol |
Array Strips Hybridization for the GeneAtlas® Instrument was carried out according to the Affymetrix protocol hybridizing array strips on the GeneAtlas® System.
Scan protocol |
Array strips were scanned using the GeneAtlas™ Imaging Station according to the Affymetrix protocol.
Description |
Gene expression data from MECs following secretory activation.
Data processing |
Raw data were GC-RMA (robust multiarray averaging) normalized, log2 transformed, and statistics performed using the Partek Genomic Suite (Partek, St. Louis, MO).
