RNA |
Source name |
9 day-old mp-B4149 seedlings, DEX/IAA-treated, replica 2
Organism |
Arabidopsis thaliana |
Characteristics |
Total seedling
Treatment protocol |
Seedlings were treated with 10 micromolar Dexamethasone during one hour in liquid MS medium, and during 1 more hour with 50 micromolar IAA.
Growth protocol |
Seedlings were grown on half-strength Murashige-Skoog (MS) media containing 1% sucrose and 0.8% Daichin Agar in a 16 hour light / 8 hour dark cycle at 23 degrees.
Extracted molecule |
total RNA |
Extraction protocol |
Purified total RNA was isolated using Qiagen RNEasy columns
Label |
biotin
Label protocol |
cRNA synthesis and labeling was done according to Schmid et al., Nature Genetics [2005] 37, 501-506
Hybridization protocol |
Hybridisation of 15ug cRNA was done overnight for 16 hours at 45ÂșC in a Hybridisation Oven 640 (Affymetrix). The protocol is conducted as described in the Genechip Expression Analysis Technical Manual, section 2 (Eukaryotic Sample and Array Processing), chapter 2 (Eukaryotic Target Hybridization) (P/N 701027, revision 5).
Scan protocol |
Arrays were scanned on an Affymetrix 3000 7G scanner, as described n the Genechip Expression Analysis Technical Manual, section 2 (Eukaryotic Sample and Array Processing), chapter 2 (Eukaryotic Arrays: Washing, Staining and Scanning (P/N 701028, revision 5).
