Total RNA of different groups of ProliHH cells was extracted using TRIzol (Invitrogen) according to the manufacturer’s instructions. RNA libraries were prepared for sequencing using standard Illumina protocols.

Data processing |
Low quality reads were trimmed. The remained reads were aligned to the human genome (Ensemble GRCh38.84) using Tophat2 FPKM (fragments per kilobase of exon per million fragments mapped) values were calculated by Cufflinks using default parameters for gene expression levels The counts expression profiles were normalized by Cuffnorms Genome_build: Ensemble GRCh38.84 Supplementary_files_format_and_content: Genes.counts-ProliHH organoids.txt: Counts for each sample (without running and mapping together with hiHep organoids)
