Treatment protocol |
Conceptuses were isolated from the mother's uterus at the embryonic day 19 (E19). Labyrinthine zone were micro-dissected under microscope from each placenta and flash-frozen in liquid N2 and stored at -80C until RNA extraction.

Extraction protocol |
Total RNA was extracted using RNeasy Midi Kits (Qiagen – 75144) and quantity and quality were verified using RNA 6000 Nano Kit (Agilent – 5067-1511) and an Agilent 2100 Bioanalyzer. Only RNA samples with RNA integrity numbers (RIN) >9.0 were used.

The total RNA was subjected to reverse transcription to cDNA using random hexamers tagged with T7 promoter and this was subsequently used as a template for cRNA amplification. The cRNA was then subjected to another round of reverse transcription followed by biotin labeling as described in the Affymetrix (Santa Clara, CA, USA) GeneChip whole transcript sense target labeling assay manual and labeled using the Affymetrix whole transcript kit according to the manufacturer's instructions

Hybridization protocol |
The biotinylated ssDNA was hybridized onto Affymetrix GeneChip MouseGene 1.0 ST arrays, which screen the expression of 28,853 genes using 770,317 probes.

Scan protocol |
Signals were detected using an Affymetrix GeneChip Scanner 3000 7G and analyzed using Partek Genomics Suite 6.4 software (St. Louis, MO, USA).

Data processing |
Array profiling was performed using the Mouse Gene 1.0 ST Array (Affymetrix) and the analysis of the data was performed using GeneSpring GX 12.1 (Agilent, Santa Clara, CA, USA), with two algorithms: RMA (Robust Multiarray Average) and PLIER (Probe Logarithmic Intensity Error).
