The median eminence from all animals within the same treatment groups are combined and single cells were dissociated via trituration in papain. Isolated cells from each treatment group were split into 2 and were encapsulated in droplets and cDNA libraries were made using a 10X Genomics Chromium instrument and 10X Single Cell 3’ V2 Reagent kit. Paired end sequencing was performed on an Illumina HiSeq 4000. The first 26bp read contains both a cell barcode and a unique molecular identifier, the next 76bp read contains the cDNA insert.
