Treatment protocol No cell lines were exposed to any type of treatment.
Growth protocol Cells were grown using standard tissue culture until 60-80% confluent at which point RNA was isolated.
Extracted molecule total RNA
Extraction protocol Total RNA was extracted from the cells using the Quick RNA MiniPrep Zymo Research Kit (Genesee scientific).
Label biotin
Label protocol The biotin-labeled cDNA was prepared from 250 ng of total RNA using the Ambion WT Expression Kit and Affymetrix WT Terminal Labeling Kit.
 
Hybridization protocol Fragmented and biotin-labeled cDNA at a concentration of 25 ng/ul or 2.25 ug total was hybridized to the Mouse Gene 2.1 ST peg plate. The Mouse Gene 2.1 ST peg plate was hybridized, washed, stained and scanned using the Affymetrix GeneTitan MC instrument. The default method for the Mouse Gene 2.1 peg plate was used on the GeneTitan. The hybridization time for the expression plates was 17 hours and the temperature was 48 C.
Scan protocol The Mouse Gene 2.1 ST peg plate was scanned using the Affymetrix GeneTitan MC instrument.

Gene expression data from 4T1-mCherry-rLuc (murine breast cancer cells labeled with mCherry fluorescent reporter and renilla luciferase) that were injected into axillary lymph nodes and then isolated from a lung metastasis on day 36 after injection
Data processing The data were analyzed using Affymetrix Expression Console and the algorithm used was RMA (with background adjustment, quantile normalization and summarization).
