Extraction protocol The cell cultures were mixed with the ice cold phenol ethanol solution (phenol: ethanol = 1: 9), and centrifuged at 16,000g for 5 min at 4°C. The pelleted cells were stored at -80°C for future use. Total RNAs were extracted using an RNeasy mini kit (QIAGEN Cat#74104), in accordance to the manufacturer’s instructions.

Data processing Basecalls performed using RTA.
