The cell cultures were mixed with the ice cold phenol ethanol solution (phenol: ethanol = 1: 9), and centrifuged at 16,000g for 5 min at 4°C. The pelleted cells were stored at -80°C for future use. Data processing |
Basecalls performed using RTA. Sequenced reads were trimmed for adaptor sequence using Trimmomatic v0.38. Trimmed reads were mapped to reference genome with Bowtie v1.1.2. Reads Per Kilobase of exon per Megabase of library size (RPKM) were calculated using HTseq v0.10.0.
