Treatment protocol |
Animals were grown until young adult stage. Then they were washed off the plates into 15 mL conicals, rinsed 2 times in S Basal, supplemented with E. coli OP50 for foos, and dispensed into 6-well plates, 6,000 worms/well. LK molecules at 100 uM final or corresponding amount of DMSO solvent control were added to wells.

Growth protocol |
N2 worms were synchronized by hypochlorite treatment. Arrested L1s were plated on 10 cm NGM plates seeded with E. coli OP50 and grown 56 h at 22°C until they reached young adult stage.

RNA was extracted using TRI Reagent (Molecular Research Center) according to manufacturer's instructions and purified using an RNeasy column (Qiagen).

Data processing |
Data were analyzed with GC-RMA in the R programming language, using the Bioconductor package gcrma. Analysis was performed using default settings.
