CHO-K1 suspension cells in 50 ml working volume in 250ml flasks (Corning) and maintained in an ISF1-X (Climo Shaker) Kuhner incubator

Extraction protocol |
A total of 4,000,000 cells were collected from the triplicate flasks grown with putrescine (+PUT) and without putrescine (NoPUT), and RNA was extracted using 1ml Trizol according to the manufacturer's instructions.

Biotinylated cRNA were prepared according to GeneChip™ WT PLUS Reagent Kit Manual Target Preparation for GeneChip™Whole Transcript (WT) Expression Arrays from 300 ng total RNA.

Hybridization protocol |
Following fragmentation, 3.5 ug of ssDNA were hybridized for 16 h, 60 rpm at 45 C on GeneChip CHO Gene 2.0 ST array. The GeneChips were washed and stained in the GenChip Fluidics Station 450.

Scan protocol |
GeneChips were scanned using the Affimetrix GeneChip scanner.
