Conidia suspension (100 ul, 5X10^6 conidia ml-1) of WT and ΔBbHapX of Beauveria bassiana were inoculated on the SDA plates (Glucose 4%, Peptone 1%, and Yeast extract 1% plus 1.5% Agar), and incubated at 25 °C for 3 days.

Extraction protocol |
Resultant mycelia of each strain were ground into powder in liquid nitrogen. Total RNA was isolated from mycelial powder using TRIzol® Reagent (Invitrogen, Carlsbad, CA, USA). The double-stranded cDNA were prepared with the Illumina gene expression sample prep kit for Illumina arrays.

Data processing |
Illumina Hiseq X Ten used for sequencing. The sequenced tags were filtered to remove low-quality tags, empty tags and tags with a copy number of <2. All the clean tags vectoring the CATG and 49 bp tag fragments were mapped to the sequenced genome of Beauveria bassiana ARSEF2860 with the precision of no more than two nucleotide mismatch. All mapped genes were quantified in terms of the expected number of fragments per kilobase of transcript sequence per millions base pairs sequenced (FPKM) with Cufflinks software. Screening of differentially expressed genes in paired library of WT/∆BbHapX. FDR (False Discovery Rate) is a method to determine the threshold of P-value in multiple tests (Benjamini, Y. and Yekutieli D. The Annals of Statistics. 2001, 29: 1165-1188). Genome_build: Beauveria bassiana ARSEF 2860; The reference genome has the accession NO. of ADAH00000000. GenBank assembly accession NO. is GCA_000280675.1. Supplementary_files_format_and_content: WT-BbHapX_Conidiation.xlsx: Excel file includes Gene ID, mean FPKM values, log2(fold change), FDR-value and gene description for the paired sample.
