Status Public on Jun 01, 2020
Title QMa-25 mJ
Sample type RNA

Source name Primary melanocytes
Organism Homo sapiens
Characteristics cell type: melanocytes
rna fraction: exosome
treatment: irradiated
Treatment protocol A dose of irradiation was applied to quiescent melanocytes on day 1 and then again 24 hrs later.
Growth protocol Cultured melanocytes were made quiescent prior to irradiation (solar UVA+UVB)
Extracted molecule total RNA
Extraction protocol By 64 hr post-irradiation, exosomes from the irradiated or un-irradiated melanocytes were subjected to RNA extraction using the Total Exosome RNA and Protein Kit (Invitrogen).
Label Fluorescence from FAM/SYBR green
Label protocol n/a

Hybridization protocol n/a
Scan protocol n/a
Description irradiated
human miRNA Card A
Data processing Raw Ct values were obtained by using Applied Biosystems TaqMan SDS v2.4 software. These values were then imported into Applied Biosystems RQ manager or Expression Suite software to quantify the relative expression of miRNAs in each of the un-irradiated and irradiated samples. Both software packages use the comparative CT method (Delta Delta CT), which translates into an RQ value.
MammU6 was used as an endogenous control, and miRNAs with a Ct value >36 or failing to amplify with an efficiency coefficient less than 1.0 were omitted from the analysis by ABI's Expression suite software.
