Treatment protocol 24h after isolation and culture, preB cells were transducted with eMLM-shRen or eMLM-shPhf23 through retroviral infection. 48h after infection, the transducted cells labelled with GFP were sorted out by FACS AriaIII. Immediately after sorting, the cells were centrifugated to obtain cell pellets, followed by Trizol addition.
Growth protocol preB cells were cultured in RPMI 1640 medium supplemented with 20% gibco FBS, 1% Pen/Strep and 10ng/ml mouse IL7.
