Treatment protocol |
The injury experiment was performed under in vivo condition for mouse and monkey eyes, and under ex vivo condition for human donor eyes. To mimic central corneal injury, an excimer laser (Allegretto Wave Eye-Q) was used to make a wounding by removing a small portion of the central epithelium and anterior stroma. The full thickness of the limbal epithelium and a layer of stroma beneath were excised to exclude the potential influence of limbal cells. Normal central corneas from contralateral eyes from the same donors without laser injury and limbal excision were used as uninjured controls.

Human, monkey and mouse single cell suspensions in 0.4% BSA-PBS were processed for single-cell sequencing using the Chromium Single Cell 3’ Reagent Version 1 Kit and Chromium Controller (10xGenomics, CA, USA). In each experiment, 20,000 cells per reaction were loaded for Gel Bead-in-Emulsion (GEM) generation in the GemCode instrument, followed by cell lysis, barcoded reverse transcription of RNA, cDNA amplification, shearing, adaptor and sample index attachment. scRNA-Seq

Data processing |
Base calling and demultiplexing was performed using Illumina bcl2fastq software. Additional cell-level demultiplexing and alignment was performed with 10x Cell Ranger software (v3.0.2). Genome_build: GRCh38/macFas5/mm10. Supplementary_files_format_and_content: Filtered feature-barcode matrices containing cellular barcodes in MEX format
