Treatment protocol Six mice were anesthetized with 1% pentobarbital sodium solution (30 mg kg-1), and unilateral hindlimb ischemia was induced by femoral artery ligation. Then, those mice were divided into two groups randomly, they were treated via intraperitoneal injection daily from days 0 to 7 with with sterile water or proanthocyanidins. Mice were deeply anesthetized with pentobarbital sodium and then ischemic gastrocnemius were collected at days 7.
Growth protocol Experimental animals were male and commercially-available C57BL/6 mice (Tengxin biotechnology co. LTD, Chongqing,China).
All mice were raised in Pathogen-free room and the room was maintained at a constant temperature of 23±1℃ with a 12 h light/dark cycle. Food and water were available ad libitum in the home cages.
Extracted molecule total RNA
Extraction protocol Total RNA was isolated using TRIzol (Invitrogen) and miRNeasy mini kit (QIAGEN) according to manufacturer’s instructions, which efficiently recovered all RNA species, including miRNAs. RNA quality and quantity was measured by using nanodrop spectrophotometer (ND-1000, Nanodrop Technologies) and RNA Integrity was determined by gel electrophoresis
Label Hy3
Label protocol The miRCURY™ Hy3™/Hy5™ Power labeling kit (Exiqon, Vedbaek, Denmark) was used according to the manufacturer’s guideline for miRNA labelling
 
Hybridization protocol The Hy3TM-labeled samples were hybridized on the miRCURYTM LNA Array (v.19.0) (Exiqon) according to array manual
Scan protocol The slides were scanned using the Axon GenePix 4000B microarray scanner (Axon Instruments, Foster City, CA).

Data processing Scanned images were then imported into GenePix Pro 6.0 software (Axon) for grid alignment and data extraction. Replicated miRNAs were averaged and miRNAs that intensities>=50 in all samples were chosen for calculating normalization factor. Expressed data were normalized using the Median normalization. After normalization, differentially expressed miRNAs with statistical significance were identified through Volcano Plot filtering
