Label protocol Commercially available Cy3-conjugated goat anti-mouse antibody was added to the microarray at 1:4000 in 5% fetal bovine serum in phosphate-buffered saline with 0.1% Tween-20.
 
Hybridization protocol Murine antiserum was probed on the antigen microarray at 1:250 dilution in 5% fetal bovine serum in phosphate-buffered saline with 0.1% Tween-20, and incubated for 1.25 hours at 4 degrees Celsius.
Scan protocol Fluorescent array images were collected for Cy3 with a GenePix 4000B fluorescent scanner at 10 micron resolution and image intensity data were extracted and analyzed with GenePix Pro 6.1 analysis software.
Description Murine antiserum was obtained from blood collected from an individual mouse of the strain R2Yaa IRF5-/-. A 1:250 dilution of this antiserum was then applied onto the Utz Lab Whole Protein Autoantigen Array (V1.0), labeled with a goat anti-mouse Ig(M + G) secondary antibody conjugated to Cy3 (Jackson ImmunoResearch), and scanned with a GenePix 4000B scanner using GenePix Pro 6.1.
Data processing After background correction and removal of flagged values, data was normalized by dividing each array value by the goat anti-mouse IgG (25 ug/ml) array feature signal intensity and then multiplying by 10,000 to obtain a normalized signal intensity value.
