Murine antiserum was probed on the antigen microarray at 1:250 dilution in 5% fetal bovine serum in phosphate-buffered saline with 0.1% Tween-20, and incubated for 1.25 hours at 4 degrees Celsius.

Fluorescent array images were collected for Cy3 with a GenePix 4000B fluorescent scanner at 10 micron resolution and image intensity data were extracted and analyzed with GenePix Pro 6.1 analysis software.

Description |
Murine antiserum was obtained from blood collected from an individual mouse of the strain R2Yaa IRF5+/-. A 1:250 dilution of this antiserum was then applied onto the Utz Lab Whole Protein Autoantigen Array (V1.0), labeled with a goat anti-mouse Ig(M + G) secondary antibody conjugated to Cy3 (Jackson ImmunoResearch), and scanned with a GenePix 4000B scanner using GenePix Pro 6.1.

Data processing |
After background correction and removal of flagged values, data was normalized by dividing each array value by the goat anti-mouse IgG (25 ug/ml) array feature signal intensity and then multiplying by 10,000 to obtain a normalized signal intensity value.
