| Extraction protocol |
Total RNA from sorted cells was extracted using the Arcturus PicoPure RNA Isolation kit (ThermoFisher Scientific, St Aubin, France) according to the manufacturer’s instructions. The RNase-Free DNase Set (Qiagen, France) was used to remove contaminating DNA. RNA integrity was assessed using an RNA 6000 Pico kit on a Bioanalyzer 2100 (Agilent Technologies).
1 to 3 ng of total RNA were amplified and converted to cDNA using SMART-Seq v4 Ultra Low Input RNA kit (Clontech). Afterwards 150 pg of amplified cDNA was used to prepare library following Nextera XT DNA kit (Illumina).
