|
Source name |
dCAM x GFAP-Cre
|
Organism |
Mus musculus |
Characteristics |
tissue: Brain striatal tissue time: 13 weeks post injection strain: Rosa26-dCas9-activator mouse (dCAM)
|
Treatment protocol |
Cells were dissected from mouse striatum (n=2). Incubation with dissociating enzyme was performed for 90 min.
|
Growth protocol |
mouse striatum. Animal housing and handling protocols were approved by the committee for the Care and Use of Laboratory animals of the Government of Upper Bavaria (Germany) and were carried out in accordance with the European Communities' Council Directive 2010/63/EU. During the work, all efforts were made to minimize animal suffering. All mouse lines were kept in a controlled pathogen free (SPF) hygiene standard environment on a 12 h light/dark cycle. The mice had access to ad libitum standard feed and water always. All tests were approved for the ethical treatment of animals by the Government of Upper Bavaria.

Extracted molecule |
total RNA |
Extraction protocol |
Chromium Single Cell 3´Library & Gel Bead Kit v2 (PN-120237, 10xGenomics) to generate single cell gel beads in emulsion (GEMs). cDNA synthesis was done according to 10x Genomics guidelines. Libraries were pooled and sequenced on a NovaSeq6000 (Illumina) according to the Chromium Single Cell v.2 specifications and with an average read depth of 50,000 aligned reads per cell.
