Sample type |
RNA |
Source name |
AF D+Q
Organism |
Mus musculus |
Characteristics |
tissue: Annulus Fibrosus genotype: C57BL/6 age: 23-month-old
Treatment protocol |
N.A.
Growth protocol |
NP and AF tissues were dissected from lumbar discs of 23M Veh (n=5) and D+Q (n=4) BL6 mice.
Extracted molecule |
total RNA |
Extraction protocol |
Pooled tissue from single animal served as an individual sample. Samples were homogenized and total RNA was extracted using RNeasy® Mini kit (Qiagen).
Label |
biotin
Label protocol |
Fragmented biotin labeled cDNA was synthesized using the GeneChip WT Plus kit according to ABI protocol (Thermo Fisher Scientific).
Hybridization protocol |
Gene chips (Mouse Clariom S) were hybridized with fragmented and biotin-labeled cDNA. Arrays were washed and stained with GeneChip hybridization wash & stain kit
Scan protocol |
scanned on an Affymetrix Gene Chip Scanner 3000 7G, using Command Console Software.
Description |
Gene expression data from 23-month-old annulus fibrosus C57BL/6 mouse treated with Dasatinib and Quercetin combination.
Data processing |
Chp files were generated by sst-rma normalization from Affymetrix cel file using Expression Console Software. Experimental group was compared with control group by using Transcriptome array console 4.0 software. The SST-RMA algorithm was used for data normalization.
