Extracted molecule genomic DNA
Extraction protocol Prior to purification, 250 mg crushed cord tissue was enzymatically digested with hyaluronidase (~2.7U in 250 µl PBS) for 30 minutes at 37 C, mixed with 250 µl TNES lysis buffer, transferred to gentleMACS M Tubes (Miltenyi 130-093-236) for 1-minute homogenization followed by overnight treatment with 200 µg Proteinase K at 55 C. QIASymphony SP was used for automated purification of (buffy coat/cord tissue) DNA in combination with QIASymphony DSP DNA Midi Kit (Qiagen #937255) and DNA was bisulfite converted using the EZ DNA Methylation Kit™ (Zymo Research).
