C. albicans strains were grown in YPD media (1% yeast extracts, 2% peptone, 2% glucose) at 30˚C. Overnigh cultured cells were diluted to OD600 of 0.1 into fresh 10ml YPD and grown at 30˚C until OD600 of 1.0.

Extraction protocol |
Total RNA was extracted using NucleoSpin® RNA (MN, MN740955) according to the protocol of the manufacturer. For sequencing, mRNA was captured using NEBNext® Poly(A) mRNA Magnetic Isolation Module (NEB, E7490). A strand-specific sequencing library was synthesized using NEBNext® UltraTM Directional RNA Library Prep Kit for Illumina (NEB, E7420) according to the instruction manual.

The Illumina Hiseq generates raw images utilizing HCS(HiSeq Control Software v2.2) for system control Base calling through an integrated primary analysis software called RTA(Real Time Analysis. v1.18) The BCL(base calls) binary is converted into FASTQ utilizing illumina package bcl2fastq(v1.8.4) Adapters are not trimmed away from the reads.
