Treatment protocol 21-day-old seedlings were transferred into the growth boxes containing 1/4 strength Hoagland’s solution with 50mmol/L NaHCO3 (pH 8.5).
Growth protocol Glycine soja seeds were grown in a culture room with 60 % relative humidity, 24℃ and a light regime of 12h light/12h dark. Germinated seedlings were grown in the growth boxes containing 1/4 strength Hoagland's solution.
Extracted molecule total RNA
Extraction protocol Total RNA was extracted from frozen leaves with TRIzol (Invitrogen, Carlsbad, CA) according to the instructions from the manufacturer.
Label biotin
Label protocol Biotinylated cRNA were prepared according to the standard Affymetrix protocol from 6 ug total RNA (Expression Analysis Technical Manual, 2001, Affymetrix).
 
Hybridization protocol Following fragmentation, 10 ug of cRNA were hybridized for 16 hr at 45℃ on GeneChip Soybean Genome Array. GeneChips were washed and stained in the Affymetrix Fluidics Station 450.
Scan protocol GeneChips were scanned using the GeneChip Scanner 3000.
Description Gene expression data from Glycine soja leaves of 21 days old after 6h NaHCO3 stress.

Data processing The computation of expression values was conducted using dChip software. Sample wise normalization to the median probe cell intensity (CEL) of all 14 arrays. Model-based gene expression was obtained from normalized CEL intensities based on a Perfect Match-only model.
