Reverse transcription was performed using the Mouse Complete SeraMir Exosome RNA Amplification and Profiling Kit (System Biosciences) according to the manufacturer's instructions. Real-time qPCR was performed (Applied Biosystems ViiA7) using standard run conditions (40 cycles at 95oC for 15 seconds and 60oC for 60 seconds.)

The normalization and all the data analysis was performed using the manufacturer's qPCR analysis templates https://systembio.com/wp-content/uploads/Mouse_miRNome_v14_2.xls For the normalization it uses the Spike-in control RNA. Target miRNA signals were normalized to the average Spike-in control RNA signals; 2^-deltaCt, where deltaCt = (Ct_Target - Ct_control average)] All deltadeltaCt based fold-change were calculated from the uploaded raw threshold cycle data automatically by the templates. Fold Change worksheet reports test/control (i.e., OCExoInhib/Vehicle) ratios.
